Genetic transformation technologies for the common dandelion, Taraxacum officinale

Abstract Background Taraxacum officinale , or the common dandelion, is a widespread perennial species recognized worldwide as lawn and garden weed. Common dandelion also cultivated for use in teas, edible greens, traditional medicine. It produces latex closely related to Russian T. kok-saghyz which being developed rubber crop. Additionally, vast majority of extant dandelions reproduce asexually through apomictically derived seeds- an important goal many major crops modern agriculture. As such, there increasing interest molecular control pathways well basic biology reproduction dandelion. Results Here we present improved Agrobacterium -based genetic transformation regeneration protocol, protocol generation protoplasts using free DNA, leaf infiltration transient gene expression. These protocols easily obtainable explants from soil-grown plants reagents most plant laboratories. We show that markers used systems function expected including fluorescent proteins, GUS, anthocyanin regulation, resistance kanamycin, Basta, hygromycin. Conclusion Reproducible, stable methods are presented will allow needed structure studies genes proteins .
